SPASTICITY-DANTROLENE

1. Peripheral site of action, reduces extrafusal & intrafusal muscle fiber activity by blocking the release of Ca 2+ from the sarcoplasmic reticulum.

2. Preferred for spasticity due to TBI, stroke, CP. Also used for malignant hyperthermia, neuroleptic malignant syndrome, Baclofen-withdrawal.

3. Adverse effects: hepatotoxicity, sedation, weakness, fatigue, paraesthesia, GIT effects.
